Abstract

Objective: Comparative study of pharmacological activity of leukomizin, austricin and badhyzin. Materials and Methods: Rats and rabbits, weighing 150-180 g and 2.5-3 kg, respectively, were used to study the influence of drugs on lipid metabolism. Both intact and rats with different hyperlipidemic states were used: starvation, ethanol and triton. In all cases of experiment, the animals received a dose of 10 mg/kg per os. Experimental atherosclerosis in rabbits was reproduced by prolonged exogenous cholesterol loading at a dose of 0.3 g/kg for 90 days. Total cholesterol, α-cholesterol, triglycerides in blood serum and tissues were determined on a biochemical analyzer. Results: In all models studied, the drugs had pronounced hypolipidemic, hypotriglyceridemic, and hypocholesterolemic effects. By all the parameters of lipid metabolism among the lactones studied, the effect of badhyzin approximated the corresponding action of lovastatin. On the model of atherosclerosis, when the rabbits were injected with lactones, especially badhyzin, the lesion area was much less pronounced; plaques were few, small and thin. The index of atherosclerotic lesion of the aorta was significantly lower. Conclusions: Among the sesquiterpene lactones of the guaiolide series studied, austricin and badhyzin have a hypolipidemic effect superior to that of leukomizin
